Identification and characterization of cross-reactive antigens from Neospora caninum and Toxoplasma gondii OAK

Murine monoclonal antibodies (mAbs) against Neospora caninum tachyzoites were produced to identify the cross-reactive antigens between N. caninum and Toxoplasma gondii. Ten mAbs recognizing cross-reactive antigens of both parasites were obtained and tentatively classified into 6 different groups based on their reactivity patterns in an indirect fluorescent antibody test and Western blot analysis. The characterization of the cross-reactive antigens of two closely related apicomplexan parasites, Neospora caninum and Toxoplasma gondii, is important to elucidate the common mechanisms of parasite–host interactions. In this context, a gene encoding N. caninum ribosomal phosphoprotein P0 (NcP0) was identified by immunoscreening of a N. caninum tachyzoite cDNA expression library with antisera from mice immunized with T. gondii tachyzoites. The NcP0 was encoded by a gene with open reading frame of 936 bp, which encoded a protein of 311 amino acids. O Toxoplasma gondii é um parasita que tem como hospedeiro definitivo todos os felídeos, e como hospedeiros intermediários os mamíferos, dentre eles o próprio felídeo, o homem e o equino. A contaminação nos herbívoros ocorre pela ingestão de gramíneas e ração contaminadas pelo oocisto. Foi atendido no Hospital Veterinário da Universidade Federal de Uberlândia um equino, macho, quarto de milha, de dois anos e meio de idade, com histórico de fezes pastosas, emagrecimento e incoordenação nos membros posteriores. Ficava com uma égua em um piquete, que morreu dez dias antes com os mesmo sinais clínicos, sendo a alimentação a base de pasto e sal mineral e água em cocho. ABSTRACT: Infections caused by protozoa belonging to the Sarcocystidae family have worldwide distribution and are common in ruminants, leading to considerable economic losses. This study evaluates Sarcocystis spp., Toxoplasma gondii and Neospora caninum infections in sheep from Southwest region of Rio Grande do Sul, Brazil. Myocardium samples of 80 sheep raised on extensive system were collected. Tissue cysts were detected by direct examination and presence of infective agents was confirmed by PCR. Aiming to improve the molecular diagnosis of symptomatic toxoplasmosis in Brazilian patients, this study evaluated the performance of real time PCR testing two primer sets (B1 and REP-529) in detecting Toxoplasma gondii DNA. Neosporosis y toxoplasmosis en la liebre europea (Lepus europaeus) en la provincia de La Pampa (Argentina) ABCL

El objetivo de este trabajo fue determinar la prevalencia a Neospora caninum (Nc) y Toxoplasma gondii (Tg) y conocer el periodo reproductivo de la liebre europea (Lepus europaeus) en la provincia de La Pampa (Argentina). Para ello se muestrearon 106 liebres procedentes de acopiadores de la provincia de La Pampa. Los sueros fueron analizados por un ensayo inmunoenzimático de competición para determinar anticuerpos a Nc y para Tg se realizó una prueba de hemaglutinación indirecta. Se encontraron 66 hembras, de las cuales 13 estaban preñadas, y 40 machos (p=0,01). Sobre un total de 44 sueros analizados para Nc, 5 (11,4%) dieron positivo. De los 106 sueros estudiados para Tg ninguno fue positivo.

Occurrence of antibodies to Neospora caninum and Toxoplasma gondii in dairy cattle from the northern region of the Paraná State, Brazil Arq. Bras. Med. Vet. Zootec.

Three-hundred and eighty-five serum samples were taken from dairy cows on 90 farms in 12 counties from the northern region of the Paraná State, Brazil. The samples were analyzed by IFAT for the detection of anti-Neospora caninum and anti-Toxoplasma gondii IgG antibodies. Forty-five (12%) samples were seropositive to N. caninum, while 102 (26%) samples were seropositive to T. gondii. Only four animals were seropositive to both coccidia. No significant difference was observed between the N. caninum serology and any of the variables studied, such as dairy cattle management, milk production, reproductive problems, feeding, and presence of dogs, cats and rodents. ABSTRACT: Lowland Tapir ( Tapirus terrestris ) is the second largest South American land mammal. It is strictly herbivorous and its exposure to Toxoplasma gondii should be indicative of environmental contamination by oocysts.In the present study antibodies to T. gondii in 47 Brazilian tapirs maintained ex situ in 10 Brazilian and in one Paraguayan institution were sought in serum samples by the modified agglutination test (MAT ≥25). Foram avaliadas, pela técnica de imunofluorescência indireta, a freqüência e a magnitude dos títulos de anticorpos antitoxoplasma (Toxoplasma gondii, Nicolle; Manceaux, 1909), em gatos infectados naturalmente pelo vírus da imunodeficiência dos felinos (VIF). Utilizaram-se 115 amostras de soro sangüíneo de gatos negativos ao vírus da leucemia felina que foram divididas em 3 grupos. Os 22 animais do grupo I eram positivos ao VIF. Os 58 animais que compuseram o grupo II eram doentes porém negativos na pesquisa de anticorpos anti-VIF e os 35 felinos do grupo III eram hígidos e negativos ao VIF. No presente trabalho, descrevem-se os resultados da detecção de anticorpos anti-T. gondii (IgG e IgM) em amostras seqüenciais de soro de cordeiros recém-nascidos e de suas mães, sem aparentes sinais clínicos da infecção. Os anticorpos IgG foram quantificados pelos testes de imunofluorescência indireta (IFI) e hemaglutinação indireta (HAI). Os anticorpos IgM foram detectados pelo método de hemaglutinação indireta 2-mercaptoetanol (HAI/ 2-ME), utilizando "kits" comerciais. Infection caused by Toxoplasma gondii, toxoplasmosis, is one of the most frequent zoonoses in the world; it normally affects both genders equally. Humans are one of several possible intermediate hosts, and the disease is oligosymptomatic in most cases. Vertical transmission is an important cause of fetal malformation and sequels in newborns. Approximately 10% of postnatal cases present multiple manifestations, ranging from low fever and mild lymphadenopathy to severe encephalitis. In moderate cases, lesions such as retinochoroiditis may emerge during acute infection or even years later. We analyzed 313 cases of toxoplasmosis from 1992 to 2004, including 261 acute cases.
